Sequoia Sciences, Inc., a drug discovery company San Diego, CA aids high-throughput screening (HTS) programs by providing novel libraries of chemically diverse compounds that it isolates from plants. The unprecedented mass sensitivity of Protasis microflow NMR probes coupled with the high-throughput, microfluidic sample manager developed expressly for use with the flowprobes, now allows Sequoia to acquire 1D proton and 2D Gradient COSY NMR spectra on quantities of less than 10 micrograms in under ten minutes in its pursuit of rapid structure elucidation.  With the aid of the ICG probe, Sequoia has been able to identify more than twenty new molecular entities (NME’s) within the last nine months, a remarkable achievement in drug discovery productivity.  The two prongs of Sequoia’s successful strategy are: (1) purification optimized by multi-stage separations methods and (2) structure elucidation using micro-scale NMR.

Sample management is one of the most critical aspects of NMR analysis. Proper preparation of the sample enables a maximum amount of information to be extracted in minimum time, and accurate control of the experiment ensures data integrity, experimental reproducibility, and offers the option of subsequent (e.g. mass spectrometry) on-line analysis. The enhanced mass sensitivity, chromatographic resolution, and concentrating ability of capillary liquid chromatography enables the application scientist to take full advantage of the enhanced NMR mass sensitivity of MRM’s Microcoil™ probes.

MRM chose to strategically align with Waters Corporation because we believe the quality of the Waters CapLC® surpasses that of any similar instrument.  Unlike split-flow systems that waste valuable deuterated solvents, the CapLC forms its solvent gradient directly with microdisplacers.  And its built-in photodiode array (PDA) UV/Vis detector includes a patented Extended Path Length flow cell to provide full sensitivity in a capillary detector flowcell.
